چکیده

Various biological and clinical applications require the quantification of the messenger RNA (mRNA) abundance of the biological materials under investigation. Reverse transcription-polymerase chain reaction (RT-PCR) is a popular technique for mRNA quantification. However, in these applications, multiple quantifications of mRNA are required from the same sample. Multiplex RT-PCR uses multiple primer pairs to amplify the specific targets simultaneously from a single sample. Therefore, multiplex RT-PCR is suitable for such applications. Complementary DNA (cDNA) microarray is a robust and versatile tool that allows researchers to examine the expression of hundreds or thousands of genes for the analysis of gene function, disease diagnosis, and drug discovery. Multiplex RT-PCR is usually performed for the cDNA preparation during the cDNA microarray experiments. Primer design is the most crucial step in multiplex RT-PCR. Therefore, the goal of this study was to implement an intelligent primer design system. The core of the system was based on mathematical techniques. The system can guide users to design cost effective primer collection. The system had also been successfully applied to two real world examples with biological and clinical significance. q 2005 Elsevier Ltd. All rights reserved.

Comparison of Two Multiplex PCR Systems for Meat Species Authentication

Background: Meat species adulteration has become a problem of concern. This study aimed to compare two previously published multiplex Polymerase Chain Reaction (PCR) methods for meat species authentication.  Methods: The primers used in the first multiplex PCR involved species-specific reverse primer for sheep, goat, cattle, pig, and donkey with universal forward primer. In the second multiple...

A Pipeline with Multiplex Reverse Transcription Polymerase Chain Reaction and Microarray for Screening of Chromosomal Translocations in Leukemia

Chromosome rearrangements and fusion genes present major portion of leukemogenesis and contribute to leukemic subtypes. It is practical and helpful to detect the fusion genes in clinic diagnosis of leukemia. Present application of reverse transcription polymerase chain reaction (RT-PCR) method to detect the fusion gene transcripts is effective, but time- and labor-consuming. To set up a simple ...

Development of a Multiplex Polymerase Chain Reaction for Differential Diagnosis of Canary Pox Virus

Background and Aims: A multiplex transcription-polymerase chain reaction (m-PCR) was developed for direct detection and discrimination between canarypox virus (CPV) and other avian poxvirus (APV). Materials and Methods: Three compatible primer sets were designed for m-PCR amplification of different loci fpv126, fpv140, and fpv167 located at highly conserved APV genes. Results: Results showed th...

Multiplex Tetra-Primer Amplification Refractory Mutation System Polymerase Chain Reaction to Genotype SNP8NRG221533 of Neuregulin-1 Gene

Schizophrenia is a severe neuropsychiatric disorder with symptoms such as hallucination, delusion and mental disorder. It is a complex disorder, in which genetic components play a crucial role in its pathogenesis. Among candidate genes for schizophrenia, Neuregulin 1 (NRG1) gene is the most important gene,  association of which with the illness has been confirmed in several studies. Single nucl...
